Summaryofprocedure: | TheTBARsassaykitmeasuresmalondialdehyde(MDA),areactivecompoundformedfromlipidperoxidesthataregeneratedunderconditionsofoxidativestress.Oxidativemodificationoflipidsoccurswithagingandvariousdiseases,andincreasedoxidativestressisassociatedwithdiabetesanditscomplications.Cellularexposuretooxidativestressgivesrisetohighlyreactiveandunstablelipidhydroperoxides.DecompositionofunstableperoxidesderivedfrompolyunsaturatedfattyacidsleadstotheMDAformation.MDAreactswiththiobarbituricacid(TBA)toformanadductthatcanbemeasuredspectrophotometricallyorfluorometrically,thelatterprovidinggreatersensitivity.DeterminationofTBARsinBIOLOGicalsampleshasbecomethepreferredmethodforassessinglipidperoxidationandoxidativestress.Exocell"sTBARsassaycanbeusedwithaspectrumofbiologicalsamplesincludingbodyfluids,tissueandcellspecimens. Tocompletetheassay,standardorsampleisreactedwiththiobarbituricacidandheatedinthepresenceofaceticacid.Aftercoolingandclarificationbycentrifugation,aliquotsareplacedintomicrotiterwellsofablackplateandreadinafluorometeratexcitation484nm,emission530nm,orintoaplasticplateforreADIngat530-540nminaspectrophotometer.MDAconcentrationinsamplesisdeterminedfromthestandardcurve. |
